Title: Subculturing and Isolation of Pure Culture
Objective:
1. To study the technique of aseptic in order to transfer microorganisms to a subculture
2. To study the technique of isolation of pure culture and perform a streak plate method
Answer: Method:
(a) Subculturing
1. The tubes to be inoculated were labelled with name and date.
2. The tubes were placed in the palm, secured with the thumb, and separated to form a V.
3. The inoculating loop was then flamed until the entire wire is red.
4. With the sterile loop in hand, the tubes were uncapped and the neck of the tubes was flamed by rapidly passing them through the flame.
5. The culture was transfer from: slant-to-broth and broth-to-slant.
6. The neck of the tubes was flamed by rapidly passing them through the flame and the tubes were recapped.
7. The loop was reflamed to sterile it.
8. All the culture was incubated at room temperature for 3 days.
(b) Isolation of pure culture (Streak plate method)
1. The agar plate was labelled with name and date
2. The side of the agar plate was flamed before opening the cover.
