10. To understand the role of the gram staining as an important differential stain.
11. To learn the usefulness of a simple stain vs. a differential stain.
Answer: Method:
(a) Preparation of bacterial smear
1. A small amount of the bacterial inoculums from the slant culture was transferred onto the center of the clean slide.
2. Then, one drop of water was took and placed on the bacterial inoculums.
3. Both of them were spread into a thin area approximately the size of a nickel.
4. The smear was quickly passed over the flame of the Bunsen burner two or three times by holding the slide at one end.
(b) Simple staining
1. The smear prepared previously from part (a) was used for the following procedure.
2. The slide was placed on the staining tray with the smear flooded with methylene blue for one to two minutes.
3. The slide was then washed with tap water to remove the excess stain. (The slide was hold in a parallel way to prevent the organisms being washed off when the stream of water flow through.)
4. The tissue paper was used to blot dry...(short extract)
