Objectives:
1. To perform a negative staining procedure.
2. To perform the spore stain procedures for differentiate between bacterial shape and vegetative cell forms.
3. To perform the capsule stain procedures to distinguish capsular material from the bacterial cell.
4. To recognize the bacterial shapes and arrangements.
5. To learn how to identify the results of the organisms stained.
Materials:
1. Petri plates containing Staphylococcus aureus, bacillus subtilis and klebsiella pneumonia
2. Inoculating loop
3. Glass slide
4. Boiling tube
5. Beaker
6. Distilled water
7. India ink
8. Malachite green
9. Safranin
10. Carbolfuchsin
11. Crystal violet
12. Acid alcohol
13. Methylene blue
Answer: Methodology:
Negative staining
1. A glass slide was prepared and one small drop of India ink was added at the side of the slide.
2. A sterilized loop was used to obtain a loopful of bacteria from the streak plate.
3. The bacteria were then mixed well with the ink and then used another new glass slide to spread the suspension with ink to form a thin smear.
4. After the slide was air dried, then the slide was observed under the microscope.
5. The observation was recorded.
Spore staining
Schaeffer-Fulton method
1. The bacteria smear was prepared first and then added with the malachite green.
2. The slide was steam-over the boiling water for 10 minutes on the small beaker.
3. After 10 minutes the slide was cooled down, and then rinsed the slide with the distilled water for 30 seconds.
4. The safranin was then added on the slide for 20 seconds.
